Structural Studies on Cytoskeletal Proteins.

چکیده

A number of componenets of cytoskeleton are very large proteins which are in their complete native form non amenable to the structure determination via X-ray diffraction techniques due to their non-globular shape and intrinsic flexibility which hinder the formation of suitable crystals. Dissecting such proteins into their representative building blocks-domains and subsequent structural studies of these have shown to give valuable structural and functional information on the biological system. Structural and evolutionary studies on actin-binding domains from spectrin and alpha-actinin as well as on the two central repeats of alpha-actinin will be presented.

A growing family: the expanding universe of the bacterial cytoskeleton.

Cytoskeletal proteins are important mediators of cellular organization in both eukaryotes and bacteria. In the past, cytoskeletal studies have largely focused on three major cytoskeletal families, namely the eukaryotic actin, tubulin, and intermediate filament (IF) proteins and their bacterial homologs MreB, FtsZ, and crescentin. However, mounting evidence suggests that these proteins represent...

The cytotoxic effect of memantine and its effect on cytoskeletal proteins expression in metastatic breast cancer cell line

Objective(s):Breast cancer is an important leading cause of death from cancer. Stathmin and tau proteins are regulators of cell motility, and their overexpression is associated with the progression and bad prognosis of breast cancer. Memantine, an N-methyl-D-aspartate (NMDA) receptor antagonist, is the potential inhibitor of tau protein in neurons. This study determines the effect of memantine ...

Genetic, biochemical and structural approaches to talin function.

The cytoskeletal protein talin plays a key role in coupling the integrin family of cell adhesion molecules to the actin cytoskeleton. In this paper I present a brief review on talin and summarize our recent studies, in which we have taken both genetic and structural approaches to further elucidate the function of the protein.

Intrinsic structural disorder in cytoskeletal proteins.

Cytoskeleton, the internal scaffold of the cell, displays an exceptional combination of stability and dynamics. It is composed of three major filamentous networks, microfilaments (actin filaments), intermediate filaments (neurofilaments), and microtubules. Together, they ensure the physical and structural stability of the cell, whereby also mediating its large-scale structural rearrangements, m...

Structural insights into bacterial modulation of the host cytoskeleton.

Many bacterial pathogens manipulate the host cell cytoskeleton during infection. Such cytoskeletal modulation can occur at several points of contact between the pathogen and the host, and involves extracellular receptors, intracellular signal transduction and cytoskeletal proteins themselves. The field of bacterial pathogenesis has progressed dramatically over the past decade, such that structu...
